GUIDE TARGET MISMATCH TOLERANCE

The figure above is from Jinek et al. 2012. The section entitled, "mismatched targets" corresponds to the noncomplementary DNA target strand. These results demonstrate that mismatch within the 5' end of the guide sequence portion of the crRNA are not tolerated well if they exceed 4 nt in length!
